Prostaglandins are bioactive signaling molecules synthesized from arachidonate by the action of cyclooxygenase (COX) isoenzymes and subsequently by PG synthase. Following formation by COX, PGH2 may be converted by PG synthase into numerous products including: PGI2, PGE2, PGD2, PGF, and thromboxane A2. Importantly, the letter designates the composition of the prostane or oxane ring; whereas the subscript number indicates the quantity of double bonds. Biologically, prostaglandins are well-known to be key mediators of the inflammatory response, and their role in the resolution of inflammation is an active area of investigation. As with other eicosanoid species, the combined efforts of lipidomic and cellular biology techniques will provide a basis for the continued identification and understanding of the role of prostaglandins in mediating health and disease and improving therapeutic design.
