Inhibitors of Phospholipase D

The main role of phospholipase D (PLD) involves the hydrolysis of membrane glycerophospholipids to phosphatidic acid. Phosphatidic acid plays a key role in signaling proteins at membrane surfaces and regulating cell survival. Phosphatidic acid also alters membrane curvature which facilitates vesicle formation leading to increased tumor cell proliferation. Inhibition of PLD1 and PLD2 has been studied as possible alternatives to hormone-based therapies to treat cancers.
