18:1(2)-16:0(16-azido, 2) Cardiolipin is a synthetic cardiolipin that incorporates two naturally occurring oleoyl (18:1) chains and two omega-azido functionalized palmitoyl (16:0) chains. This clickable lipid provides a versatile bioorthogonal handle for the post-formulation functionalization of liposomes and lipid membranes. The azide moiety readily undergoes highly specific CuAAC or SPAAC with alkyne-tagged molecules. This clickable cardiolipin is particularly useful for preparing labeled cardiolipin in model membranes and mitochondrial membrane mimetics, and for studying cardiolipin-dependent membrane organization and lipid–protein interactions while minimizing perturbation during the initial formulation step.
