GD2 Cer (d18:1/16:0) is a complex, glycosphingolipid featuring a highly polar oligosaccharide headgroup and a ceramide backbone composed of a sphingosine (d18:1) base and saturated fatty acid. While normally restricted to low expression levels in the central nervous system and peripheral nerves, GD2 is prominently overexpressed on the surface of tumors of neuroectodermal origin. The unique tissue distribution of this material makes it a premier tumor-associated antigen and critical target for cancer immunotherapy, including monoclonal antibody treatments.
Beyond clinical applications, GD2 plays a fundamental role in cell-to-cell adhesion, signal transduction, and the formation of functional lipid rafts on the plasma membrane.
