GDN A85525 (Legacy code 850525) glyco-diosgenin (GDN)
Glyco-diosgenin (GDN) is a fully synthetic, steroid-based glycoside detergent designed as a modern alternative to digitonin in membrane protein research. Built around a hydrophobic diosgenin (steroidal sapogenin) core and a hydrophilic di-glucose head group, GDN delivers the gentle solubilization properties researchers expect from digitonin while addressing many of the limitations associated with the naturally derived detergent.
Because digitonin is isolated from Digitalis purpurea (foxglove), its composition can change between production batches and may contain trace cardiac glycosides such as digitoxin and digoxin. Additionally, digitonin solutions are prone to clouding and reduced stability over time, which can interfere with their use in biophysical methods (such as isothermal titration calorimetry and surface plasmon resonance) by increasing the background signal or obstructing instrumentation.
Researchers who use GDN instead don’t have to worry about these issues because it is produced through a fully synthetic manufacturing process, providing reliable batch-to-batch consistency, improved water solubility (>10% at 20°C vs. digitonin), and a lower critical micelle concentration (~18 µM).
Why GDN Over Digitonin or DDM?
GDN offers a compelling balance of performance and stability. Its mild detergent properties help maintain native protein-lipid interactions and fragile multiprotein assemblies, making it well-suited for structural biology workflows.
Unlike digitonin, GDN doesn’t have issues with limited solubility, cloud formation, or variability inherent in plant-derived materials. In comparison to stronger detergents, such as DDM (n-dodecyl-β-D-maltoside), GDN is often preferred when maintaining complex stability—particularly for cryo-electron microscopy (cryo-EM) studies—is the primary objective over maximum extraction efficiency.

Application
GDN is widely used in structural biology to solubilize and stabilize membrane proteins before cryo-EM analysis. Its combination of mild detergent activity, excellent solution stability, and consistent composition makes it a trusted replacement for digitonin in many research laboratories.
The detergent has supported a range of published cryo-EM structures, including investigations of GPCR-G protein complexes and other membrane-associated protein assemblies, where preserving native interactions throughout purification was essential for successful structure determination.
